RESUMEN

Pump-probe experiments at X-ray free-electron laser (XFEL) facilities are a powerful tool for studying dynamics at ultrafast and longer timescales. Observing the dynamics in diverse scientific cases requires optical laser systems with a wide range of wavelength, flexible pulse sequences and different pulse durations, especially in the pump source. Here, the pump-probe instrumentation available for measurements at the Single Particles, Clusters, and Biomolecules and Serial Femtosecond Crystallography (SPB/SFX) instrument of the European XFEL is reported. The temporal and spatial stability of this instrumentation is also presented.

RESUMEN

The cold tolerance of winter rapeseed (Brassica napus) cultivars is critically important for winter survival and yield formation in northern China. Few studies have examined the genetic mechanism underlying the overwintering survival of B. napus. Here, an F2 population including 174 lines and an F2:3 population including 174 lines were generated to identify the quantitative trait loci (QTLs) related to the cold tolerance of B. napus. A genetic linkage map including 1,017 markers merged into 268 bins covering 793.53 cM was constructed. A total of 16 QTLs for two cold-tolerance indicators related to overwintering success were detected among the two populations. These QTLs were responsible for explaining 0.97%-12.74% of the phenotypic variation. Two major QTLs, qOWRTA07 and qOWRLA07, explaining more than 10% of the phenotypic variation were identified in overlapping regions, and we suspected that these two QTLs might represent the same QTL mapped between the two bins, c07b004 and c07b005, corresponding to the physical interval from 21.4 M to 23.4 M on chromosome A07. One gene, BnaA07G0198300ZS, contained the candidate region for overwintering rate (OWR). RT-qPCR analysis showed that the expression of this gene significantly differed between the two parents (NST57 and CY12), and its expression was higher in NST57 than in CY12. This gene may be involved in the cold-response during overwintering period of B. napus. These results are important for the molecular breeding to improve the cold tolerance and overwintering success of winter oilseed rape.

Sclerotinia stem rot, caused by Sclerotinia sclerotiorum, is a devastating disease in rapeseed. The objective of this study was to investigate the role and the mechanism of silicon (Si) in alleviating the disease severity of S. sclerotiorum in rapeseed. In the absorption assays, the rapeseed that absorbed 10 mM of K2SiO3 exhibited an 86% decrease in lesion size on infected leaves as compared with controls. In the spray assay, the lesion length on rapeseed stems was reduced by 30.5-32.9% with the use of 100 mM of a foliar Si fertilizer as compared with controls. In the pot assay, the lesion length on rapeseed stems was reduced by 34.9-38.3% when using the Si fertilizer as basal fertilizer. In the field assay, both the disease incidence and disease index of sclerotinia stem rot were significantly reduced with the usage of a solid Si fertilizer, Si foliar fertilizer, and the application of both, without negative affection on the main agronomic traits and seed quality of rapeseed. The transcriptome sequencing, quantitative reverse transcription PCR (qRT-PCR), and biochemical assays between K2SO4- and K2SiO3- treated rapeseed leaves revealed that Si promoted the biosynthesis of defense-related substances and enhanced the antioxidation and detoxification abilities of rapeseed after infection. Thus, this study concluded that Si can alleviate the disease severity of S. sclerotiorum in rapeseeds, partially due to the induced defense responses.

RESUMEN

User operation at the European X-ray Free-Electron Laser Facility started at the SASE1 undulator beamline in fall 2017. The majority of the experiments utilize optical lasers (mostly ultrafast) for pump-probe-type measurements in combination with X-ray pulses. This manuscript describes the purpose-developed pump-probe laser system as installed at SASE1, implemented features and plans for further upgrades.
